To determine the viable cell count of a bacterial culture, you have plated 50 µL of a 100-fold diluted sample of the culture on a nutrient agar plate and obtained 20 colonies after overnight incubation. The viable cell count of the culture is ______ CFU mL^-1. (answer in integer)
GATE 2026 BT Q.33: solution and answer
Answer and solution
ANSWER
The viable cell count of the culture is 40000 CFU per mL.
Official answer: 40000Source: Official answer key, GATE 2026 BT (IIT Guwahati)
Each colony grows from one viable cell, so the count of colony forming units (CFU) on the plate equals the number of viable cells in the volume plated. The viable count of the original culture is .
Convert the plated volume to mL: .
Concentration of the diluted sample: .
The sample was diluted 100-fold, so the original culture is 100 times more concentrated: .
The viable cell count of the culture is , that is 40000 as an integer.
KEY IDEA
Viable count = colonies / volume plated (in mL) x dilution factor. Convert the volume to mL first, then multiply back by the dilution to get the concentration of the original culture.
WATCH OUT FOR
Forgetting to convert 50 microlitres to 0.05 mL (giving 20/50 = 0.4 and then 40), or forgetting to multiply by the dilution factor and stopping at 400 CFU per mL.
