Answer and solution

ANSWER

(B) Myoglobin (17 kDa). In gel filtration, proteins come out in order of decreasing size: Albumin (66 kDa), then Thrombin (37 kDa), then Myoglobin (17 kDa), then Insulin (6 kDa).

Official answer: BSource: Official answer key, GATE 2026 BT (IIT Guwahati)

  1. How gel filtration (size-exclusion chromatography) works: the column is packed with porous beads. Small molecules can enter the pores, so they have more volume to pass through and come out later. Large molecules are kept out of the pores, travel only through the space between the beads, and come out earlier. So the elution volume (the volume of buffer collected before a protein comes out) goes down as molecular mass goes up.

  2. For native globular proteins inside the column's fractionation range, size follows molecular mass , and a calibrated column gives the empirical relation , where and are column constants. The heaviest protein therefore has the smallest and comes out first.

  3. Sort the four proteins by molecular mass, largest first: Albumin () > Thrombin () > Myoglobin () > Insulin ().

  4. So they elute in this order: 1st Albumin, 2nd Thrombin, 3rd Myoglobin, 4th Insulin.

  5. The protein that elutes third is Myoglobin (), option (B).

KEY IDEA

Gel filtration separates by size. Large molecules are kept out of the bead pores and come out first. Small molecules enter the pores and come out last. For globular proteins, falls roughly linearly with , so the elution order is simply the order of decreasing molecular mass.

WATCH OUT FOR

Getting the elution order backwards. In ion-exchange or affinity chromatography, a molecule is held back by binding. In gel filtration nothing binds: the small proteins are the ones slowed down, because they wander through the pores. Students who think big molecules are 'held up' pick Thrombin (C) as the third to elute. That is the second protein counting from the small end.
