GATE 2025 · BTQ.62 · 2 marks · NAT

A 2 L bioreactor is being operated as a chemostat, at a flow rate of 0.8 L/h and sterile feed of 10 g/L substrate. The bacterial growth follows Monod kinetics at a maximum specific growth rate of 0.6 h with a Monod constant of 0.5 g/L and a biomass yield coefficient of 0.4 g/g.

The exit biomass concentration is _______ g/L.

Answer and solution

ANSWER

The exit biomass concentration is 3.6 g/L.

Official answer: 3.4 to 3.8Source: Official answer key, GATE 2025 BT (IIT Roorkee)

  1. Dilution rate, where is the volumetric flow rate and the working volume: .

  2. At steady state in a chemostat with sterile feed (), and with no cell death or maintenance, the biomass balance with gives specific growth rate = dilution rate: .

  3. Monod kinetics: , where is the maximum specific growth rate, is the Monod constant and is the substrate concentration in the reactor (which equals the exit concentration in a well-mixed chemostat). Setting and solving for :

  4. Washout check, with the feed substrate concentration: . Since , a steady state without washout exists.

  5. Steady-state substrate balance, with the biomass yield on substrate (grams of cells formed per gram of substrate consumed):

  6. Exit biomass concentration .

KEY IDEA

At steady state in a chemostat with sterile feed, the specific growth rate equals the dilution rate (). Putting that into the Monod equation gives the substrate concentration in the reactor, . The yield then gives the biomass, . Check first that is below the washout dilution rate.

WATCH OUT FOR

Taking as the flow rate itself (0.8 h), which is above and so wrongly predicts washout. Another is using the feed concentration in place of and getting g/L. It is also possible to swap and when rearranging the Monod equation for .
