GATE 2024 · BTQ.60 · 2 marks · NAT

A fed batch process is running at quasi-steady state with respect to substrate and biomass concentration. At 2 h, the culture volume is 500 L with a constant sterile inlet feed at 50 L h of glucose. The culture kinetic parameters and are 0.2 h and 0.1 g L, respectively.

The substrate concentration in the reactor will be ________ g L (rounded off to one decimal place).

Answer and solution

ANSWER

The substrate concentration in the reactor is 0.1 g L^-1.

  1. At quasi-steady state in a fed-batch culture, the total biomass keeps rising but the biomass concentration and the substrate concentration stay constant. Growth has to exactly offset the dilution caused by the incoming feed, so the specific growth rate equals the instantaneous dilution rate: , where is the feed flow rate and is the culture volume.

  2. Dilution rate at , where : .

  3. Monod kinetics, with the maximum specific growth rate and the saturation constant (the substrate concentration at which ): .

  4. Set and solve for : , so

  5. Substitute:

  6. Quick check: here , so by the definition of the answer has to be . The feed glucose concentration is not given and is not needed. At quasi-steady state it sets the biomass level (, with the biomass yield on substrate), not .

KEY IDEA

At quasi-steady state in a fed-batch reactor, , using the volume at that moment. Putting this into Monod kinetics gives , the same result as a chemostat but with a dilution rate that falls as the volume grows.

WATCH OUT FOR

Using the starting volume or a time-averaged volume instead of the volume at the stated moment (500 L at 2 h). Another mistake is thinking the feed glucose concentration is needed: it fixes the biomass concentration, not the residual substrate. Some students also swap numerator and denominator and write .
